在活的非凋亡细胞中，RPE-模联蛋白V检测非诱导细胞中的先天性细胞凋亡，其通常为所以细胞的2-6%。在凋亡细胞中，RPE-模联蛋白V与磷脂酰丝氨酸结合，磷脂酰丝氨酸位于细胞膜的外叶上，因此导致染色强度增加。

图1.使用Cell MeterRPE-膜联蛋白V结合细胞凋亡检测试剂盒检测RPE-膜联蛋白V对Jurkat细胞中磷脂酰丝氨酸的结合活性。 将Jurkat细胞在37℃，5％CO2培养箱中在没有（蓝色）或1μM星形孢菌素（红色）的情况下处理4-5小时，然后用RPE-膜联蛋白V染色30分钟。 使用FACSCalibur（Becton Dickinson）流式细胞仪使用FL2通道测量RPE-膜联蛋白V的荧光强度。
